A Karyogram obtained from a culture of my(=Junko Suzuki) peripheral blood, showing a normal female karyotype. The procedure is described in the following text:

A culture was set up in RPMI 1640 medium, supplemented with 15% fetal bovine serum, phytohemagglutinin, and an aliquot of Junko Suzuki's peripheral blood. After incubation for 72 hours at 37 degrees centigrade, metaphase cells were collected by a short-term treatment of Colcemid. The cells were processed to hypotonic treatment with 0.075M KCl, then fixed with methanol/acetic acid mixture(3:1). The cell suspension was dropped onto clean, chilled slide glasses and air-dried. The slides were treated with trypsin and stained with Giemsa. Well spread metaphase cells were photographed microscopically at a high-power magnification.
